I. Background
白細(xì)胞介素 23 （IL23）是慢性炎癥性疾病（如銀屑病、炎癥性腸病、多發(fā)性硬化癥或類風(fēng)濕性關(guān)節(jié)炎）發(fā)展的關(guān)鍵促炎細(xì)胞因子，與 IL12、IL27、IL35 和 IL39 一起屬于 IL12 家族。過度 IL23 信號轉(zhuǎn)導(dǎo)的病理后果與其促進(jìn)靶細(xì)胞群（主要是分泌 Th17 和 IL17 的 TCRγδ 細(xì)胞（Tγδ17））產(chǎn)生炎癥介質(zhì)的能力有關(guān)，例如 IL17、IL22、粒細(xì)胞-巨噬細(xì) 胞集落刺激（GM-CSF）或腫瘤壞死因子（TNFα）。這些致病介質(zhì)促進(jìn)粒細(xì)胞和巨噬細(xì) 胞的募集和活化，導(dǎo)致組織損傷，誘發(fā)慢性炎癥，最終發(fā)展出臨床癥狀。 IL23 是一種異二聚體細(xì)胞因子，由兩個二硫鍵連接的結(jié)合亞基 p40（由 IL12β編碼）和 p19 （IL23α）形成，它們與 IL12Rβ1 的 IL23R 形成的膜受體復(fù)合物結(jié)合。IL23 與 IL12 共享 p40 亞基及其受體成分之一 IL12Rβ1。盡管存在相同的亞基，但 IL23 和 IL12 具有不同的生物學(xué)功能。IL23 刺激 JAK2 和 TYK2 活性，導(dǎo)致信號轉(zhuǎn)導(dǎo)和轉(zhuǎn)錄激活因子（STAT）的激活。
