| I. Background | |
整合素αvβ6 是由αv 和β6 亞基組成的異二聚體跨膜受體,屬于整合素家族成員,其胞 外域通過識(shí)別含 RGD(天冬氨酸)序列的配體介導(dǎo)細(xì)胞與細(xì)胞外基質(zhì)的相 互作用,主要在上皮細(xì)胞中低表達(dá),但在組織損傷、纖維化、炎癥及惡性腫瘤中顯著上調(diào), 成為疾病治療的潛在靶點(diǎn)。該整合素的主要配體包括細(xì)胞外基質(zhì)蛋白如纖維連接蛋白 (Fibronectin)、層粘連蛋白(Laminin)以及潛活形式的 TGF-β,后者通過與αvβ6 結(jié)合被 激活為活性形式,驅(qū)動(dòng)纖維化和免疫抑制微環(huán)境形成;此外,αvβ6 還可作為口蹄疫病毒 (FMDV)等病原體的入侵受體。 | |
